PURPOSE:To carry out the regeneration of actinomycetes protoplast in high regeneration efficiency, by using a protoplast regeneration medium having a particular composition containing alkali metal succinate or sucrose, etc. and having a double-layered structure. CONSTITUTION:A double-layered medium is prepared by using (A) a soild flat plate of a protoplast regeneration agar medium containing (i) alkali metal succinate or sucross, (ii) a magnesium salt and calcium salt at higher concentrations than the salt in the conventional medium, (iii) agar or agarose, and (iv) other components for conventional regeneration medium, as the lower layer, and (B) a medium containing an alkali metal succinate or sucrose, a magnesium salt and a calcium salt at high concentration, soft agar or agarose, and other conventional components, as the upper layer. Protoplast of actinomycetes is cultured in the upper layer and/or at the boundary part of the upper layer and the lower layer. The surface of the upper layer is dried before cultivation. |
