The invention relates to process for preparing 5'-inosinic acid (IMP) from a carbon source, hypoxanthine (Hyp), and a phosphoric acid donating material using a microorganism as an enzyme source. In this process, use is made of both a microorganism having the activity of yielding 5-phosphoribosyl-1-pyrophosphoric acid (PRPP) or its precursor from a carbon source and another microorganism having the activity of yielding IMP from both of Hyp or its precursor and PRPP or its precursor.
