2050925 9011355 PCTABS00002 A method of introducing foreign nucleic acid, especially a gene into birds, especially chickens, takes advantage of the developmental stage of the embryo into which primordial germ cells (PGCs) collect in the germinal crescent, migrate in the blood stream and settle in the germinal ridge, which becomes the gonad. The method comprises providing in vitro foreign nucleic acid which it is desired to introduce into the germ line of a bird, introducing the foreign nucleic acid into explanted primordial germ cells of a bird and then introducing these primordial germ cells into the blood system of a recipient embryo of an incubated egg, at a stage of embryonic development at which introduced primordial germ cells will settle in the germinal ridge.
