PURPOSE:To enable efficient obtaining of 1,2-dehydrosteroids in high yield by preparing 1,2-saturated-3-ketosteroids in the form of an aqueous suspension using a water-soluble polymer dispersing agent and then bringing the resultant suspension into contact with microbial cells, etc., of a specific microorganism. CONSTITUTION:A microorganism of the genus Arthrobacter is cultured in a culture medium containing glucose, etc., and about 0.005wt./vol.% 1,2- dehydrated-3-ketosteroid compound (A) expressed by formula I (R is alkyl) to provide microbial cells (treated substances) (B) having steroid-1-dehydrogenase activity. The resultant component (A) in an amount of >=5mg/ml is then mixed with an organic solvent, such as toluene, and a water-soluble polymer dispersing agent, such as polyvinyl alcohol, at 0.01-1 weight ratio of the component (A) to afford an aqueous suspension (C). The component (B) is subsequently added to the component (C) so as to provide about 1g/l concentration thereof and reacted at 20-50 deg.C and pH5-10 for 2-144hr while being stirred with aeration to afford a reaction product, which is then extracted with ethyl acetate, etc., to produce 1,2-dehydrosteroids expressed by formula II (R1 is H or COR2, provided that R2 is the sam alkyl as R in formula I). |
