A process for producing B virus surface antigen (I) by recombinant DNA techniques comprises (a) culturing saccharomyces cerevisiae 20B- 12 (II) in YEPD medium at 30 deg. C for 18 hrs, (b) precipitating the cultured (II) by centrifuge and suspending in 0.1 M LiAc soln., (c) adding the pMH 148 and pMH 149 plasmid to the suspended soln. and leaving it at 30 deg. C for 1 hrs., (d) heating the left soln. at 42 deg. C for 5 min and treating it by zymolase, (e) precipitating it by centrifuge and dissolving the precipitate in 50 mM phosphate buffer (pH 7.2) contg. 0.01% triton x-100, and (g) obtaining the supertanant by centrifuge.
