PURPOSE:To obtain immunosuppressive agent, etc., in high yield by culturing specific strain of genus Streptomyces in nutrient medium and recovering from the medium. CONSTITUTION:FR-901154 substance-producing strain and/or FR-901155 substance-producing strain is cultured in a medium containing glucose, etc., at 20-40 deg.C for 50-250 hour in stirring with aeration to obtain cultured substance. Next, said cultured substance is purified by centrifuge and extraction, etc., to collect FR-901154 substance having physical and chemical properties such as: shape:white powder; mass spectrum: by FAB-MS, m/z=812(M+Na); molecular formula: C43H67NO12; color reaction: positive to H2SO4, etc., negative for ninhydrin reaction, etc., solubility: soluble in methanol, etc., insoluble in water; melting point: 96-97 deg.C; specific optical rotation: [alpha] =-65 deg. (C=1.7, CHCl3); ultraviolet absorption spectrum: terminal absorption, etc., and chemical structure expressed by the formula. Then, the aimed immunosuppressive agent is produced containing said FR-901154 substance as active ingredient. |
