The invention relates to a process for labelling a nucleic acid in a nucleic probe comprising a nucleic-probe sequence capable of hybridizing with a complementary nucleic-target sequence, process in which the labelling of the nucleic acid is performed by direct chemical coupling of an indicator system, in particular an indicator enzyme, by means of a heterobifunctional compound.
