PERFORIN GENE

摘要

NEW MATERIAL:A perforin gene consisting of DNA sequence shown by the figure. USE:Preparation of recombinant perforin. PREPARATION:Perforin is purified from a mouse killer cell strain and an amino acid in an N end range is determined by protein sequencer using Adman degradation method. Several kinds of oligonucelotides having a base sequence complementary with the amino acid are synthesized, a clone having cDNA complementary with a prove is screened from cDNA library of mouse killer cell by using the oligonucleotides as the prove and perforin cDNA is isolated.
