PURPOSE:To enable advantageous measurement of a basic fibroblastic cell growth factor (bFGF) by fusing a splenic cell of a mammal immunized with the bFGF to a homologous or heterologous lymphocytic cell. CONSTITUTION:A splenic cell of a mouse immunized with a bFGF which is a polypeptide containing an amino acid sequence expressed by the formula is subjected to cell fusion with a lymphocytic cell that is myeloma to provide a hybridoma, which is then cloned to afford (A) a cloned hybridoma. The resultant cell (A) is subsequently propagated in a liquid culture medium or mammalian abdominal cavity to provide (B) a propagation liquid component, which is purified by salting out, etc., to produce a monoclonal antibody having the following properties and specifically binding to the bFGF. Molecular weight; 140-160 kdaltons. Without cross-reacting with acidic fibroblastic cell growth factors. Immunoglobuglin class; belonging to IgM or IgG, etc. The above- mentioned antibody, as necessary, is used to detect and determine the bFGF.
|
