PURPOSE:To highly purify the title enzyme precursor by making a urokinase-like enzyme precursor produced in bacteria such as recombinant Escherichia coli adsorb on an alkyl group-linked polymer gel followed by fractional eluation of said precursor. CONSTITUTION:Firstly, a urokinase-like enzyme precursor is produced through bacteria such as recombinant Escherichia coli incorporated with the gene of urokinase-like enzyme precursor. Thence, this urokinase-like enzyme precursor is made to adsorb on an alkyl group-linked polymer gel followed by fractional eluation to regulate the concentration of the amine-based organic solvent in the eluted solution, thus purifying the urokinase-like enzyme precursor. For said alkyl group-linked polymer gel, the alkyl group is of 1-10C, the polymer gel being of e.g. cellulose as a natural polymer; said polymer is used through suspension in water and packing into a conventional column. |
