6alpha- or 6beta-substd. oestradiol derivs. of formula (I) are new: where R = OH, NHR" or NHR"; R' = a labelling gp.; R" = the residue of a carrier protein. Antibodies obtained by immunising an animal with (I; R = NHR") in the presence of an adjuvant, opt. alternately with another immunogen, are also new. Competetive-binding immunoassays for a hapten (Hap) are effected using: (a) a labelling conjugate of the type Hap-B1-X and an antibody obtained by immunisation with a conjugate of the type Hap-B2-Y1 where B1 and B2 are different covalent linking gps. bonded to Hap in the same posn., X is a labelling gp. and Y is a carrier protein; (b) a labelled conjugate of the type Hap-B3-X and an antibody obtained by immunisation with a conjugate of the type Hap-B3-Y, where B3 is a covalent linking gp. bonded to Hap in the same posn. but in different isomeric configurations in the two conjugates; or (c) a labelled conjugate of the type Hap-B4-X and an antibody obtained by immunisation of the same animal alternately with conjugates of the type Hap-B5-Y and Hap-B6-Y, where B4 is a covalent linking gp., and B5 and B6 are different covalent linking gps. bonded to Hap in the same posn. In all cases B1-B6 must be bonded to Hap in a posn. that does not affect the specific recognition sites of Hap. USE/ADVANTAGE - The immunoassays are eps. useful for determn. of oestradiol. The various modifications are designed to improve sensitivity by minimising prodn. of antobodies specific to the linking gp.
