PURPOSE:To obtain the title factor in high efficiency by culture, in a nutrient culture medium, microorganisms belonging to the Escherichia having in their cells a recombinant plasmid capable of manifestation of human nerve growth factor-fused protein. CONSTITUTION:A recombinant plasmid containing human nerve growth factor gene segments having a DNA sequence of the formula I (Y represents from the second base of first terminated codon to a restriction enzyme split end, 3'-end; Y' is the complementary sequence of Y, representing to restriction enzyme split end, 5'-end), capable of self-growth in microorganisms and of manifesting human nerve growth factor-fused protein is incorporated into the microorganisms belonging to the Escherichia. Thence, said microorganisms is put to culture in a nutrient culture medium to accumulate human nerve growth factor-fused protein, which is then collected and digested with thrombin, thus obtaining the human nerve growth factor. |
