COUNTING OF SPERMATID AND RATIO OF LIVING SPERMATID OR FAST DETERMINATION METHOD AND APPARATUS FOR EITHER THEREOF

PURPOSE: To realize a method for evaluating sperm easily and inexpensively at high rate and high accuracy satisfying the productional requirements by employing a solution of fluorochroming iodide propidium as a buffer. CONSTITUTION: When a sample 11 encased in a cuvet is set in a cuvet holder 2, emission intensity of sample is indicated automatically by a detection electronic unit 10. Emission intensity F0 is measured for a buffer dissolving fluorochroming iodide propidium and then the intensity F1 is measured after adding a sperm sample to the solution. Subsequently, a cell membrane permeable chemicals is added thereto and the intensity F2 is measured, followed by measurement of intensity F3 after adding another buffer and permeable chemicals thereto. Finally, a sperm sample is added to the buffer and the intensity F4 is measured. Concentration of spermatid can be calculated according to expressing [α. (F2 -F3 )-(F3 -F4 )}/F0 ] and the ratio of survival spermatid can be calculated according to expression II [100-100×(F1 -F0 )-(F3 -F4 )}/(F2 -F3 )-(F3 -F4 )].
