NEW MATERIAL:A glycoprotein having cardiac action, isolated from Conus striatus, having about 25,000 molecular weight calibrated by gel filtration, about 22,000 molecular weight calibrated by gel electrophoresis, about 4.7 isoelectric point by density gradient electrophoresis, showing a single band by electrophoresis of sodium dodecylsulfate-polyacrylamide. USE:Showing cardiac actions on vertebrate animals, being expected to be useful as a cardiac. PROCESS:Conus striatus preserved in a frozen state is thawed, poison glands are taken out and venom is squeezed with keeping it at about 4 deg.C, and crude venom is collected. It is extracted with a phosphate buffer solution (pH7.0), the extracted solution is subjected to gel filtration by a column with Sephadex G-100, the prepared active fraction is treated by straight line gradient method using a column with DEAE-Sephacel, and finally it is subjected to gel filtration using a column with Sephadex G-100, to give an active fraction with a single peak. |
