<p>NEW MATERIAL:A physiologically active substance, produced by a human lymphoblastic cell HO-323 and capable of promoting antibody production of a hybridoma. USE:A reagent for promoting production of a human type monoclonal antibody. PREPARATION:For example, a human lymphoblastic cell HO-323 is suspended in a culture medium containing 10% bovine fetal blood serum and cultivated in the presence of 5% CO2 at 37 deg.C for 2 days. Cells are then separated by centrifugation to provide a cultivation supernatant, which is subsequently subjected to column chromatography. The obtained eluate is then ultrafiltered through various ultrafiltration membranes to afford various fractions, which are subsequently dialyzed to examine the activity of promoting antibody production using a human-human.hybridoma. Thereby the aimed antibody production promoting factor is obtained from the resultant fractions having high activity.</p> |
