PRODUCTION OF USEFUL GENE PRODUCT BY FILTRATION CULTURE

PURPOSE:To readily and inexpensively obtain a gene product useful for mass production, by cultivating a specific transformed host cell in a medium containing phosphoric acid by a culture device of filtration type, filtering and removing phosphoric acid and recultivating. CONSTITUTION:A structural gene which manifests control of a promoter of an alkali phosphatase gene and codes the same (different) useful protein is bonded to the downstream of the promoter to give plasmid pYK269. Then Escherichia coli is transformed with the plasmid to give E.Coli MC1061/pYK269. Then the transformant is cultivated at a first stage of given time by a culture device of filtration type consisting of a culture tank 1 storing a medium 2 containing phosphoric acid, a filter 11 made of ceramic for cross flow filtration and a level controller 24 and phosphoric acid in the medium 2 is filtered and removed by the filter 2. Then a phosphoric acid-free medium is fed from a feed tank 26 to the tank 1 and culture is carried out at second stage to give a useful gene product such as beta-galactosidase.
