PURPOSE:To obtain a high-reliability index for diabetes by bringing a sample of red cells in blood, etc. into contact with a dissociating material contg. the condensation product of phosphoric acid, etc., to dissociate the unstable type hemoglobin A1C contained in the red cells, etc., to obtain the hemoglobin and measuring the stable type hemoglobin A1C. CONSTITUTION:The sample contg. at least the red cells and/or hemoglobin in the blood is brought into contact with a medium for dissociation contg. the condensation product of the phosphoric acid and/or the salt of the condensation product of the phosphoric acid to dissociate the unstable type hemoglobin A1C contained in the red cells and/or the hemoglobin to the hemoglobin and glucose. The contact time of the sample and the dissociating agent vary with the kind, concn. pH conditions, etc. in this case and is usually 10-30min at a room temp. The sample is thereafter subjected to ion exchange chromatography, by which the respective components of the stable type hemoglobin A1C are separated, eluted and measured. The high-reliability index for diabetes is thereby obtd.
