PURPOSE:To readily detect a protein coding gene, by forming a vector having replication initiation codon, ampicillin-resistant gene, etc., and a multicloning site containing breakage sites of specific restriction enzymes. CONSTITUTION:Three vectors having replication initiation codon, ampicillin- resistant gene, beta-galactosidase, etc., and a multicloning site containing breakage sites of restriction enzymes HindIII, PstI, SalI, etc., are formed. A desired vector is selected from the three vectors, the vector and a protein coding gene are cleft by given restriction enzymes and both are bonded. Then the protein coding gene is detected by discriminating change of color. The three vectors contain frames corresponding amino acids at breakage sites. |
