PURPOSE:To obtain a liposome containing a liposomal membrane component substance therein without deteriorating a thermally unstable drug and using an organic solvent, by heating the above-mentioned component substance and a small amount of an aqueous solution to a specific temperature, cooling both, then adding a drug and aqueous solvent and treating the resultant blend with ultrasonic waves. CONSTITUTION:A liposomal membrane component substance, e.g. a membrane component substance obtained by adding a membrane stabilizer (e.g. cholesterol), charged substance (e.g. stearylamine) and antioxidant to phospholipid and hydrogenated substance thereof, and a small amount of an aqueous solution are heated above the phase transition temperature of the above-mentioned membrane component substance, gelatinized and cooled. A drug to be contained therein and aqueous solvent are then added and the resultant blend is treated with ultrasonic waves to form the aimed liposome. The initial aqueous solution is used in an amount of preferably 0.5-50pts.wt., especially 1-10pts.wt. based on 1pt.wt. liposomal membrane component. |
