PURPOSE:To determine reduced form nicotinamide with a high accuracy, by reacting a reduced form nicotinamide coenzyme-containing sample liquid, containing a nonionic surfactant and adjusted to pH4-7 with a tetrazolium based colorant. CONSTITUTION:A nonionic surfactant in an amount of 0.3-10wt.% is added to a sample liquid containing reduced form nicotinamide coenzyme, such as reduced form nicotinamide adenine dinucleotide or reduced from nicotinamide adenine dinucleotide phosphate, and the pH thereof is simultaneously adjusted to 4-7. The resultant sample liquid is subsequently reacted with a tetrazolium based colorant to determine the reduced form nicotinamide coenzyme by the coloring degree thereof. The concentration of the reduced from nicotinamide coenzyme in the sample liquid is preferably about 0.001-50mM and the pH of the sample liquid is preferably adjusted by a method for using a buffer solution.
