PURPOSE:To count blood corpuscles at a high speed highly accurately, by adding the fluorescent label body of an antibody, which is diathetically combined with a special kind of the blood corpuscles in blood, into said blood, flowing said blood in a flow cell, projecting linearly polarized light on the blood, and measuring fluorescent anisotropy ratio of the combined body of the fluorescent label antibody and the blood corpuscles. CONSTITUTION:An antibody, which is diathetically combined with blood corpuscles to be measured is formed, in subclassified counting of the blood corpuscles such as normocytes, reticulocytes, and lymphocytes in blood. A fluorescent material is attached to said antibody. Said fluorescent label antibody is added to a sample blood, and a combined body with the blood corpuscles to be measured is formed. Said blood 25 is flowed into a flow cell 23 together with a sheath liquid 24. Light from a linearly polarized laser oscillator 21 is projected. The face that the polarization anisotropy ratio of said combined body is larger than the polarization anisotropy ratio of uncombined label antibody is utilized. Then, the anisotropy ratio is detected by using an optical system, which measures the polarized light from the oscillator 21 and the polarized light which is perpendicular to said polarized light. Thus the blood corpuscles are counted at a high speed highly accurately. A wavelength selecting filter is added to the optical system, and many kinds of different corpuscles can be simultaneously counted. |
