PURPOSE:To achieve a higher analysis accuracy in a reaction of a sow speed and a multi-stage reaction, by inserting a non-liquid permeable film detachably between a reagent layer and a development layer. CONSTITUTION:Reagent layers 1 and 2, a non-liquid permeable film 4 and a development layer 3 are laminated sequentially to produce a multi-layer analysis element. The film 4 is provided with a removing handle 40. For example, the reagent layer 1 is a regenerative cellulose film which contains glucose, glucose oxidase, 4-antiaminopyrine and 1,7-dihydroxy naphtharene, the reagent layer 2 a microfilter with modified human IgG immobilized and the development layer 3 a regenerative cellulose film. A mixed solution of an anti-human IgG labelled by peroxidase and a human serum (sample) are spotted on the multi- layer analysis element to react for 30min. Thereafter, the non-liquid permeable film 4 is removed to measure absorbance of reflected light on the side of the reagent layer 1.
