<p>PURPOSE:To make saffron stigma available all the year round, by using a sprouting saffron stigma of a stage of from about 60 days to about 2 days before blooming after the actualization of the presence of the stigma and carrying out tissue culture of the stigma in a specific medium. CONSTITUTION:Murashige-Skoog medium is added with plant hormones consisting of 0.1-20ppm of NAA and 0-20ppm of BA (excluding the combination of 0.1ppm of NAA and 0-0.1ppm of BA). A sprouting saffron stigma of a stage of from about 60 days to about 2 days before blooming after the actualization of the presence of the stigma is subjected to tissue culture in the obtained medium. As an alternative method, a Murashige-Skoog medium added with plant hormones consisting of a combination of 10ppm of 2,4-D and 0.1-10ppm of BA may be used as the culture medium. The culture of the stigma by the above method induces a callus, redifferentiates the callus and forms a stigma on the surface of the callus.</p> |
