PURPOSE:To separate and determine accurately glandular kallikrein by allowing a trypsin inhibiter to interact with a mixture of glandular kallikrein and plasma kallikrein and deactivating the plasma kallikrein. CONSTITUTION:A trypsin inhibiter is allowed to interact with a mixture of glandular kallikrein obtd. from a pancreas and plasma kallikrein in plasma and serum to deactivate the plasma kallikrein. Then, a substrate which fluoresces due to the decomposition using glandular kallikrein, e.g. the substrate shown by a formula is added to measure 440nm fluorescence of 7-amino-4-methylcoumarin isolated from the substrate. The glandular kallikrein injected as a vasodilator is distinguished from the plasma kallikrein in plasma, serum and lymph, and can be determined selectively and accurately. |
