PURPOSE:To obtain the titled enzyme having excellent heat resistance and storage stability and suitable for an alcohol sensor in good efficiency, by cultivating a yeast belonging to the genus Torulopsis in a nutrient medium. CONSTITUTION:The genus Torulopsis yeast such as Torulopsis nagoyaensis M-2003 strain (FERM P-9130), etc., is cultivated in a medium containing preferably methanol as the sole carbon source and nitrogen source, phosphate, magnesium salt, metal ion, trace element, etc., as others at 25-37 deg.C and 4.5-6.5pH for 48-72hr to isolate the aimed enzyme from cultivated product. The enzyme catalyzes reaction in which an alcohol is oxidized to give an aldehyde and hydrogen peroxide and has remaining activity of about 90% when the enzyme having about 6-8.5 optimum pH, about 6.0-8.0 stable pH, about 35-40 deg.C optimum temperature and about 5.7X10<5> (gel filtration method) is treated at 50 deg.C for 15min. |
