NEW MATERIAL:A thermostable protease NCA1503 having the following characteristics. Molecular weight; 26,000 [sodium dodecyl sulfate (SDS)-(polyacrylamide gel electrophoresis (PAGE)]. Thermal stability; Without substantially exhibiting deterioration of activity by heat treatment at 80-90 deg.C for 30min. pH activity; Suitable pH range of proteolytic activity is 6-7 in using casein as a substrate with the optimum pH of 7.0-7.5. Optimum temperature; Optimum temperature of proteolytic activity is 35-80 deg.C in using casein as a substrate with the optimum temperature of 70 deg.C. Inhibition; Inhibited by EDTA without inhibited by phenyl-methyl-sulfonyl-fluoride. USE:An enzyme stable not only to heat but also to chemicals. PREPARATION:A bacterium belonging to the genus Bacillus, e.g. Bacillus stearothermophilus NCA1503 (FERM-P No.8978), etc., is cultivated in a culture medium containing 15-20% yeast extract and 1.5-2.5mM calcium chloride. |
