PLANT GENE HAVING CARBONIC ANHYDRASE PRODUCING CODE, PLASMID AND USE THEREOF

PURPOSE:To produce the titled enzyme, by preparing cDNA complementary to mRNA of the titled enzyme extracted from green alga Chlamydomonas and introducing a plasmid in which the above-mentioned DNA inserted into a host such as Escherichia coli or the like. CONSTITUTION:A carbonic anhydrase is induced in a cell of green alga Chlamydomonas after moving from growth condition having high CO2 concentration to low CO2 condition and so the total RNA is prepared from the above- mentioned cell and a cell in non-induction phase. RNA having poly(A) is separated from the non-induction cell using mAP paper and cDNA is synthesized from the above-mentioned RNA by a reverse transcriptase. The above-mentioned cDNA is crossed with a poly(A)<+>RNA obtained from the cell in induction phase to provide a mRNA selectively induced only in induction phase. A double- stranded cDNA is synthesized from the above-mentioned mRNA by the reverse transcriptase using plasmid vector DNA, etc., of Escherichia coli. The Escherichia coli is transformed and the strain producing plant carbonic anhydrase activity is selected and recombinant plasmid having the aimed DNA is selected.
