A vaccine capable of proucing T cell-dependent antibody to H. influenza type B polysaccharide is prepd. by mixing a C4-8 dicarboxylic acid (II) and hydrazide derivatised diphtheria or tetanus toxoid in cyanogen halide-free soln. with a cyanogen halide activated capsular H. influenza B polysaccaride (III). The conjugate (I) produced has a mol. wt. of 140,000-4,500,000 and a ribose/ protein ratio of 0.25-1.0. (III) is previously sized by heating unitl over 60% is of mol. wt. 200,000-2,000,000 (dextran std., by gel permeation chromatog.) and comprises equimolar amts. of ribose, ribitol and phosphate. (I) is esp. useful for prevention of Haemophilus infection in small children and also useful for immunisation against diphtheria and tetanus.
