PURPOSE:To provide a more practical method for the determination of a tumor marker, by determining the titled substance containing a sugar nucleotide composed of unlabeled sugar and a substance bonding exclusively to a sugar transfer acceptor produced by the transfer of said sugar. CONSTITUTION:A buffer solution containing a sugar transferase to be determined is added with an unlabeled sugar nucleotide, a sugar protein labeled with <125>I and having a terminal to be subjected to transfer and bonding, beads having immobilized lectin having specificity to the transfer bonded sugar, necessary metallic ion and an activation agent at prescribed concentrations. The solution is incubated usually at about 37 deg.C for several min - several hr, the beads are washed with a buffer solution and the radiation activity is measured by a gamma-counter. The sugar nucleotide is e.g. UDP-sugar, CMP-sugar, CDP-sugar, etc., and the acceptor is e.g. monosaccharide, oligosaccharide, sugar protein, glycolipid, etc. The terminal sugar is determined by the sugar transferase.
