PRODUCTION OF RECOMBINANT VACCINIA VIRUS

PURPOSE:To obtain a recombinant vaccinia virus, by preidentifying a DNA range not essential for multiplication of viccinia virus and using the identified range for a recombination site. CONSTITUTION:DNA having promoter function and adventitious DNA capable of manifesting under the control of the DNA are integrated into a DNA range not essential for multiplication of vaccinia virus to give a recombinant vaccinia virus. In the operation, if DNA to code an enzyme capable of readily detecting enzymatic activity is used as the adventitious DNA, a nonessential DNA range of vaccinia virus genome can be readily searched and identified by a simple operation and yet selection of recombinant can be easily carried out by using the DNA range as a recombination site.
