<p>PURPOSE:To produce a monoclonal antibody useful for the diagnosis of autoimmune diseases, by fusing a tumor cell with a spleen cell of a mouse immunized with human factor I and culturing the obtained hybridoma. CONSTITUTION:A Balb/c mouse is immunized thrice with purified human factor I and Freund's complete adjuvant and spleen is extracted from the immunized mouse. The spleen cell is fused with a mouse myeloma cell strain (P3-X63-Ag8.653) originated from Balb/c mouse and resistant to 8-azaguanine and the fused cell is cloned to select a hybridoma strain producing anti-human factor I monoclonal antibodies 3-2-3, 1-3-3, 1-1-1, 1-5-3, 3-6-1 and 2-5-2. A monoclonal antibody can be produced by proliferating the above hybridoma in abdominal cavity of a mouse and the obtained antibody has high specificity to human factor I and is utilizable for the determination of said factor.</p> |
