PURPOSE:To efficiently obtain the titled compound useful for a reagent to measure alpha-amylase, by acting a specific nitro compound and silver oxide or hexadecaacetylmaltopentaosyl bromide, reacting and deacetylating the resultant product with an amine. CONSTITUTION:Hexadecaacetylmaltopentaosyl bromide expressed by formula I (Ac represents acetyl) is reacted with a compound expressed by formula II (X represents Cl or H) (example; 4-nitrophenol, etc.) and silver oxide in the presence of acetonitrile. The obtained beta-(4-nitro- or 2-chloro-4-nitro-phenyl)- hexadecaacetylmaltopentaoside expressed by formula III is reacted and deacetylated with an amine (example; ammonia, triethylamine, etc.) in the presence of water and methanol to give the aimed compound expressed by formula IV.
