QUANTITATIVE ANALYSIS OF COMPLEMENT

PURPOSE:To easily and efficiently quantize the complement, by a method wherein a bio-specimen is used as it is without dilution to be subjected to the antigen- antibody reaction with a plurality of kinds of monoclonal antihuman complement antibodies and the formed antigen-antibody composite is irradiated with light to measure photochemical change quantity. CONSTITUTION:A bio-specimen such as a serum specimen is used as it is without dilution to be subjected to the antigen-antibody reaction with two or more kinds of monoclonal antihuman complement antibodies. The formed antigen- antibody composite is irradiated with light and the change quantity of light scattering intensity or transmitted light is measured to quantize the complement. Said monoclonal antihuman complement antibody is produced by the hybridoma formed by the fusion of the cell from the tumor line of a mouse and the splenic cell of a mouse preliminarily immunized with the human complement C3 or C4.
