发明名称 METHOD FOR SEPARATING SMALL-SIZED CYCLIC DOUBLE-STRANDED SATELLITE DNA IN PROCARYOTIC CELL PARTICLE
摘要 PURPOSE:To easily obtain a small-sized cyclic double-stranded DNA without necessitating complicate operation, by using filtration process in place of centrifugal separation process. CONSTITUTION:A suspension of procaryotic cell particles is filtered to collect the procaryotic cell particles on a filter. The collected particles are washed and immersed in a lyzing liquid to destruct the procaryotic cell and decompose protein included in the cell. A solution having high salt concentration is added to the liquid to precipitate impurities, which are separated by filtration. The filtrate is added with ethanol to precipitate small-sized cyclic double-stranded DNA and the supernatant liquid is removed. A small-sized cyclic double-stranded satellite DNA such as replicative form of plasmid or virus can be separated and collected by this process from procaryotic cell or procaryotic cell particle such as mitochondria of eucaryotic cell or chloroplast.
