CYCLIC DOUBLE-STRANDED DNA, MICROORGANISM CONTAINING SAID DNA AND PRODUCTION OF POLIOVIRUS CAPSID PROTEIN P1 USING SAME

摘要

PURPOSE:A bacterium in Escherichiae transformed with a plasma containing a specific cyclic double-stranded DNA is cultured to collect the fused protein accumulated whereby poliovirus capsid protein P1, which is used as a vaccine, is obtained. CONSTITUTION:A plasmid containing 3 DNA regions coding (1) high manifestation promoter operator and the amino-terminal moiety in the m-pyrocatechol gene containing the SD sequence of the pyrocatechol gene which follows the operator and is affected by it, (2) the DNA part which follows the above region and coding the capsid protein of attenuated poliovirus Sabin 1, 3 or 3 strain and (3) a part coding the anpicillin resistance is prepared. A bacterium in Escherichia is transformed with the plasmid and cultured in a nutrient medium. Then, the fused protein accumulated in the cell bodies is collected to obtain the objective poliovirus capsid protein P1.
