DNA FRAGMENT CONTAINING CITRIC ACID SYNTHASE-PRODUCING GENE

PURPOSE:To carry out transformation of a glutamic acid-producing coryne-type bacteria or a strain producing valuable substance by genetic engineering, by separating a DNA fragment containing citric acid synthase-producing gene from a glutamic acid-producing coryne-type bacteria. CONSTITUTION:(1) Whole DNA is extracted from a cell of a glutamic acid- producing coryne-type bacterial strain and cut with a restriction enzyme. (2) A vector DNA is cut and cleaved with a restriction enzyme. (3) The DNA fragment prepared in the step (1) is integrated to the cleaved site of the vector DNA to obtain a cyclized recombinant DNA. (4) The recombinant DNA is introduced into a host E.coli. (5) A strain having desired gene is selected and separated from the E.coli containing introduced recombinant DNA.
