PURPOSE:To make quick quantitative determination with high accuracy by using a method for dissolving a sample prepd. by pulverizing toad cake (Bufonis Venenum) or toad cake (Bufonis Venenum)-contg. material with a specific concn. of a acetonitrile as an extraction solvent, subjecting such soln. to an ultrasonic treatment and injecting the soln. into a mobile phase in a reversed phase mode. CONSTITUTION:The acetonitrile having 40-80% concn. is added as the extraction solvent to the sample prepd. by pulverizing the toad cake (Bufonic Venenum) or toad cake (Bufonis Venenum)-contg. material and further a flavone is added as an internal standard material thereto, then the soln. is subjected to the ultrasonic treatment for >=10min. A prescribed amt. of the acetonitrile having the same concn. is added to such soln. to prepare the sample soln. The sample soln. is subjected to liquid chromatography in the reversed phase mode by an HPLC method (high-performance liquid chromatography) to obtain the chromatogram for the three kinds; bufalin, cinobufagin and rsibufogenine which are the essential components of bufadienolide. The respective components are easily, quickly and accurately analyzed by the above-mentioned method.
