A novel alkaloid compound speciofoline, being a crystalline substance of empirical formula C22H28N2O5, m. pt. 202-204 DEG C., optical rotation [a ]22 DEG = - 103 DEG (2% in CHCI3), D U.V. absorption maxima lambda max. 223 mm (log e =4.47), 242 mm (log e -4.27) and 290 mm (log e =3.49), minima lambda min. 230 mm (log e =4.08), 274 mm (log e =3.14); i.r. absorption bands (in hydrocarbon oil) at 2500, 1700 (broad), 1620, 1600, 1280, 1260 and 1230 (doublet), 1145, 1102, 995, 808, 780 and 750 cm.-1 and having an HBr salt of m.pt. 219-221 DEG C., is extracted from the leaves of Mitragyna speciosa by extracting the powdered dried leaves with ethanol, evaporating, dissolving the resulting syrup in glacial acetic acid, diluting, removing the precipitate formed and washing with 3% acetic acid, combining the acetic acid liquors, adding ammonia and treating with oxalic acid to precipitate the alkaloid oxalates; dissolving the oxalates in 3% acetic acid, making alkaline with ammonia, extracting with ether, adsorbing the ether extract on alumina, eluting successively with ether to give the alkaloid mitragynine followed by chloroform, evaporating the chloroform extract, dissolving the residue obtained in 5% sulphuric acid, purifying with charcoal, adding ammonia, extracting with ether and evaporating to give the desired speciofoline. The product may be converted to its pharmaceutically acceptable salts. Pharmaceutical preparations having analgetic and anti-tussive activity comprise the above speciofoline or its acid addition salts and a carrier, suitably in a form adapted to oral or parenteral administration, e.g. tablets, trochees, lozenges or capsules.
