PURPOSE:Yolk phospholipid is fractionated, and the phosphatidylethanolamine fraction is further fractionated to give the titled compounds, especially arachidonic acid (AA) and docosahexaenoic acid (DHA) from inexpensive and readily available substance through simple operations. CONSTITUTION:Yolk phospholipid is subjected to adsorption-partition chromatography to obtain phosphatidylethanolamine (PE) and phosphatidylcholine (PC). PE is subjected to alcoholysis with tetrabutylammonium hydroxide in methanol to obtain a fatty acid ester mixture of e.g., methyl esters of highly unsaturated fatty acids in which AA and DHA are highly concentrated. Then, the product is treated with a urea adduct to remove saturated acids and subjected to adsorption chromatography impregnated with silver nitrate or reversed phase partition chromatography (the latter is preferred where DHA is eluted first, then AA is eluted) to collect AA and DHA, separately.
