PURPOSE:To easily fractionate and measure the Bdelta (delta bilirubin) and Bc (conjugate type bilirubin) existing in the body fluid of a living body by measuring the coloration of an azo dye formed by the reaction of the bilirubin and diazonium salt. CONSTITUTION:a) A sample and bilirubin oxidase are first incubated in the presence of a buffer having pH ranging 9-11 to erase the Bc contained in the sample. The unreacted Bdelta and Bu are, therefore, contained in the resultant mixture of reaction. b): The resultant mixture of reaction formed in a) and the diazonium salt are mixed to from the azo dye. The intensity of the coloration of the formed azo dye is proportional to the concn. of Bdelta. c): The sample and the diazonium salt are mixed and the coloration of the azo dye formed in accordance with the reaction of DB (direct type bilirubin) and the diazonium salt is measured. d): The measured value obtd. in b) is substrated from the measured value obtd. in c), by which Bc is calculated.
