Allergy-suppressing factor (ASF) is obtd. by (a) preparing hybridomas of human T cells able to express ASF, by (i) isolating human peripheral blood lymphocytes and cultivating them for 7 days, (ii) fusing with a clone of cellular Jurkat JKOT line resistant to thioguanine and ouabaine and susceptible to hypoxanthine, aminopterine and timidine (HAT) by means of polyethylene glycol, and (iii) cultivating the F cells; (b) isolating the hybridomas by cultivation of individual clones in presence of penicillin and streptomycin, inserting the culture in a human atmos. with 5% CO2 for 7 weeks in presence or absence of HAT; and (c) collecting the ASF produced from the supernatant liq. of the culture.
