发明名称 METHOD OF PURIFYING ERYTHROPOIETIN
摘要 PURPOSE:To obtain in high yield the titled high-purity substance useful as a hematinic without damaging biological activity, by subjecting partially purified erythopoietin to reversed phase high performance liquidchromatography and then to high performance molecular sieve chromatography. CONSTITUTION:Firstly, partially purified erythropoietin is adsorbed on a porous carrier (preferably silicate type octadecyl cyanate having 200-350Angstrom pore diame ter) packed into a column of reversed phase high performance liquid chromatography and eluted with a solution of a guanidine salt in a lower alco hol by concentration gradient elution method. Then, the fraction is subjected to high performance molecular sieve chromatography to give the aimed sub stance. A column having preferably <=100,000 scavenging limit of protein molecu lar weight is preferable as a column for the high performance molecular sieve chromatrography and its eluting solution is preferably a buffer solution which contains 0.1-0.2 M NaCl and is at 7-8 pH.
