<p>PURPOSE:A mechanism to prevent free flotation and transfer of plant cell is set in a medium, a colony is formed by liquid shaking culture which is advantageous with respect to multiplication rate for the culture of colony selection of plant cell and the colony selection is carried out. CONSTITUTION:The agar medium 2 having a great number of holes having 0.5-1 mm diameter and 1-2mm depth on the surface is set at the bottom of a culture container, a cell to be cultivated is plated on the agar medium and the cells are put in the holes. The liquid medium 1 is softly poured on it and shaking culture is carried out by the operation of the shaker 3. Period of colony formation can be shortened.</p> |
