PURPOSE:To achieve a highly accurate assay of prostaglandin, by a method wherein a reagent is added to a biological composition or a body liquor to stabilize prostaglandin immediately after it is sampled, a stabilized derivative is absorbed on a filler in a fine column and after the adsorbed derivative is eluted to attach the derivative eluted to a liquid chromatograph. CONSTITUTION:Immediately after a biological tissue or a body liquor is sampled or immediately after the thawing thereof in several days once frozen at -20 deg.C immediately, a 0.1% methanol tetrahydro-furan mixed solution of 9-anthryl diazomethane is added thereto to stabilize prostaglandin in the sample in a reaction at the room temperature for 1hr and the supernatant of the solution is extracted by centrifugal separation. The supernatant is made to pass through a fine column 1 filled with a silica opposite phase based filler 2 or the like to absorb a stabilized derivative. The derivative absorbed is eluted by CHC l3 or the like, which is removed from the elution liquid passing an N2 gas therethrough and then, the derivative evaporated dry is dissolved into alcohol. The solution is analyzed by a liquid chromatograph thereby enabling the detec tion of prostaglandin in a picogram order by measuring the intensity at the excitation wavelength of 365nm and the fluorescence wavelength of 412nm.
