<p>IgE is identified and quantified by first treating test serum with an allergen (I) attached to an insoluble support. The support is then incubated with anti-IgE antibody (A6), pref. monoclonal, labelled with a fluorogenic enzyme. Unconjugated A6 is removed and the support incubated with a substrate soln. which produces a fluorescent prod. in presence of the enzyme. The fluorescence of the resulting soln. is measured. Esp. the support is an opaque microtitre plate made of polystyrene or poly(styrene-vinyl monomer) and most pref. it attached to a conjugate of (I) with a soluble protein or amino acid polymer (which improves affinity of (I) for the support). Claimed (I) are pollen, moulds, smut, animal dander or epidermal cells, insects, insect venom, dust and foods. The method is sufficiently specific and accurate for use in identifying the cause of allergies and in optimisation of desensitising dose regimes.</p> |
