PURPOSE:To provide a novel protoplast hybrid cell having the drug component (antitumor active component) of Fomes japonicus and the proliferability and edibility of Pleurotus octreatus, and obtained by the cell fusion of Fomes japonicus and Pleurotus octreatus. CONSTITUTION:The pieces of the tissue of fruit body of Fomes japonicus and Pleurotus octreatus are implanted aseptically in Hamad's culture base for Armillaria edodes to effect the growth of mycelia. The mycelia are proliferated, transplanted in Hamada's liquid media for Armillaria edodes, subcultured, and treated with an enzyme liquid to separate the protoplast of respective fungi. The separated protoplast is filtered with a filter, and centrifuged at 700xg. The residue is added with 0.5-0.7M aqueous solution of NaCl, and the dispersion and centrifugal separation procedures are repeated three times. The protoplast suspensions prepared by the above process are mixed together and centrifuged. The residue is dispersed in 30% PEG solution at 30 deg.C, cultured, added with a sufficient amount of an aqueous solution of NaCl, and centrifuged to obtain the hybrid cell of Fomes japonicus and Pleurotus octreatus. |
