PURPOSE:To facilitate the detection and measurement of an antinucleus antibody by combining a specified artificial polynucleosome insolving antigen and a labelled reagent in which an enzyme is bound to a second antibody or protein A that specifically reacts with an anti-artificial nucleus antigen antibody to be measured instead of doing therewith to make a reagent. CONSTITUTION:About 50mul of an about 2mugml refined double-strand DNA solution is bonded to a plastic carrier on a flat-bottomed micro titer plate and about 50mul of a tris sodium chloride solution containing a histon subgroup other than H1 on an equipmolar basis is added thereto to react and after the mixture is left alone at the room temperature for about 2 hours, DNA left unreacted and histon are washed away thoroughly by TBS. Then, for example, when using alkaline phosphatase-proteion A bond as labelled reagent, a non-antigen adsorbed portion on the plate is shielded by about 100mul of a TBS solution containing bovaine serum albumin (approx. 0.01%) and then, after left alone for 1hr, the product is washed away by the TBS solution containing 0.05% of Tween-20 to obtain an insolving artificial nucleus antigen. |
