PURPOSE:To enable the determination of base sequence even in a minute quantity of a specimen by enhancing electrophoretic separation capacity, in a gel electrophoretic apparatus, by forming a gel into a circular form and electrophoretic nucleic acid fragments from the outer periphery toward the center. CONSTITUTION:An electrophoretic apparatus is constituted by interposing a gel, for example, a polyacrylamide gel between two upper and lower discs 1, 2 and rotated around a center shaft 8. Then, a specimen comprising nucleic acid fragments cut at regions of bases having to receive MW separation in various lengths is injected from an injection port 4. A high voltage DC electric field is formed between the ring shaped cathode 5 provided to the outer periphery of the electrophoretic layer and the anode 6 provided to the central part thereof and the specimen is allowed to perform electrophoresis from the outer periphery toward the center through fan shaped electrophoretic zones 11. This electrophoretic specimen is irradiated with light by detectors 9, 10 and fluorescence is generated in the labelled specimen to be detected by said detectors 9, 10. Therefore, nuleic acid fragments are conc. at every electrophoretic zone during phoresis and the base sequence of a nucleic acid can be determined even in a minute amount of a specimen and the apparatus can be miniaturized. |
