PURPOSE:To measure a specified deoxyribonucleic acid (DNA) and ribonucleic acid (RNA) handily and accurately, by binding two kinds of enzymes to one- strand polynucleotide will be hybridized with one-strand polynucleotide to be measured. CONSTITUTION:One-strand polynucleotide to be measured contains DNA and RNA. Enzyme 1 and enzyme 2 are selected in such a combination that material generated from the action of the enzyme 1 constitutes the substrate of the enzyme 2. Then, the enzyme 1 (or an affinitive substance thereof) and the enzyme 2 (or an affinitive substance thereof) are bound to one-strand polynucleotide as labelled polynucleodie. So, this labelled polynucleotide is brought into contact with the polynucleotide being measured to form a two-strand polynucleotide, to which an enzyme for limiting two-strand polynucleotide is given and acts thereon to cut the two-strand polynucleotide and then, the reaction of the enzyme 2 bonded to the polynucleotide is measured. This can eliminate such a troublesome procedure as immobilizing the polynucleotide being measured to the solid phase. |
