PURPOSE:To fabricate an immunity sensor having high specificity without losing the physiological activity of protein, by crosslinking the protein of an antibody or an antigen under a mild condition by a crosslinking agent and immobilizing the protein on the surface of an electrode. CONSTITUTION:As an antibody to be immobilized, one containing no impurities, for example, a monoclonal antibody prepared by a cell fusion process, concretely, human serum albumin(HSA), adenosine-3,5'-cyclic phosphate of hapten is used. As a crosslinking agent, glutaraldehyde or bis-diazobenzene is used. For example, HSA and glutaraldehyde are added to a buffer solution and applied to or adsorbed by an electrode material, for example, glass or platinum and crosslinked to immobilize HSA on the surface of the electrode material to fabricate an immunity sensor. By this sensor, the potential of an antigen or antibody solution is measured. Because immobilization is performed by using the crosslinking agent, the immunity sensor can be simply prepared and a minute amount of an antigen and an antibody can be measured. |
